My wife and I were at a coworker's house for a dinner party. My wife spoke without being addressed or spoken to first. This is obviously unacceptable. What would be an appropriate punishment?

what makes it so wrong for your wife to speak to someone without being addressed or spoken to first? She was just talking. It's not some horrible crime for a women to start a conversation or start talking first. She did nothing wrong and It's not unacceptable for her to start a casual conversation, deal with it...
Force her to play backgammon for 1 week straight with very few breaks. After 80 hours of backgammon she will be a backgammon master and you can win money from tournaments. With the money you can raise a family (only reproduce boys, God willing) and buy a significant house in the suburbs.
