Slope is rise over run. Run is horizontal distance, left to right. Run is always positive because we always go left to right. Rise is the vertical change in that same distance. A negative rise means it drops.
The first line rises -2 units from -2 to 0 and runs 6 units from -4 to 2 so the slope is -2/6 = -1/3. Any line perpendicular has to have the negative reciprocal slope, which is +3. The second line rises -6 units from 8 to 2 and runs -1 unit from 3 to 2 so the slope is 1/6. It is not perpendicular.
