This answer focuses more on the ethics of the issue. I believe that if the patient is willing to participate in the experiment and has been informed of ALL the possible side effects, it should be allowed based on one of the four pillars of medical ethics; autonomy. The drug should also have gone through extensive testing to detect any negative side effects, and if any are found then the trials should be stopped.
Also, the effects of that drug on certain patients may be different on others, for example a chemical imbalance or some other condition may affect what the drug does, and how it works.
I'm not quite sure what you mean by how it links to those things you mention though.
