No gene is resposible for ageing, rather all genes are. What happens is that when cells divide they get "capped" with a telemerase protein which works as a counting mechanism to prevent a cell being copied to many times as that introduces defects into the process. It is this process that causes ageing and eventually enough cells are unable to duplcate themselves successfully that the organism dies.
We DO regenerate our cells, our body has replaced every cell at least once every 7 years on average, but the regeneration process is not perfect, imperfections creep in and the biological process to prevent catastrophic failure leads causes ageing and eventually death.
