I'm applying to grad school and a question I'm being asked to answer in an essay format is who is my most important role model and how this individual has influenced my development. My role model is honestly a fictional character. (Commander Shepard from Mass Effect if you're wondering). Would this be the kiss of death if I were to write about him and not a real person?

No. In my opinion either it is real person or a fictional character, it does not really matter.. We are influenced by the character and it does not depend if it is real or not.. Very often we like some film actors or starts and they can also influence on our mind and behaviour.. However they are not real people in real life.. they play fictional characters.
I do not agree with the previous answers. However, you need to be really careful with your idea, you may write about anybody you like. All what really matters if the essay is structured good and if all your paragraphs are coherent.
